The discovery of efficacious new human therapeutic agents is one of humanity’s most vital
tasks. It is an enormously demanding activity that requires creativity, a vast range of scien-
tific knowledge, and great persistence. It is also an exceedingly expensive activity. In an
ideal world, no education would be complete without some exposure to the ways in which
new medicines are discovered and developed. For those young people interested in science
or medicine, such knowledge is arguably mandatory.
In this book, Douglas Johnson, Jie Jack Li, and their colleagues present a glimpse
into the realities and demands of drug discovery. It is both penetrating and authoritative.
The intended audience, practitioners and students of medicinal and synthetic chemistry,
can gain perspective, wisdom, and valuable factual knowledge from this volume. The
first two chapters of the book provide a clear view of the many complexities of drug dis-
covery, the numerous stringent requirements that any potential therapeutic molecule must
meet, the challenges and approaches involved in finding molecular structures that “hit” a
biological target, and the many facets of chemical synthesis that connect initial small-scale
laboratory synthesis with the evolution of a process for successful commercial production.
The remaining 15 chapters provide a wealth of interesting synthetic chemistry as applied
to the real world of the molecular medicine of cancer, infectious, cardiovascular, and
metabolic diseases. At the same time, each of these chapters illuminates the way in
which a first-generation therapeutic agent is refined and improved by the application of
medicinal chemistry to the discovery of second- and third-generation medicines.
The authors have produced a valuable work for which they deserve much credit. It is
another step in the odyssey of drug finders; a hardy breed that accepts the high-risk nature
of their prospecting task, the uncertainties at the frontier, and the need for good fortune, as
well as focus and sustained hard work. My ability to predict the future is no better than that
of others, but I think it is possible that a highly productive age of medicinal discovery lies
ahead, for three reasons: (1) the discovery of numerous important new targets for effective
disease therapy, (2) the increasing power of high-throughput screening and bio-target
structure-guided drug design in identifying lead molecules, and (3) the ever-increasing
sophistication of synthetic and computational chemistry.

pher's stone of crank cookery: ''The Fester Formula" - an
invention that is to amphetamines what breadmaking ma-
chines are to a freshly baked loaf of bread. What is normally
a time-consuming, laborious and somewhat messy operation
is reduced to a sort of "meth in a box." Don't be surprised to

The DEA has recently estimated the total number of clandestine
LSD labs operating in the United States at only 100, with most of
them located in northern California. This alarmingly low number of
labs leaves the supply of LSD in this country at constant peril. Further,

lysergic acid or lysergamide are controlled substances and unavailable

n 1991 an unusual book with an unusual title first began to circulate
among an unusual set of people: those convinced that there were significant
psychological and spiritual benefits to be derived from the use of psyche-
delic drugs. PIHKAL: A Chemical Love Story, consisted of two parts: Book I
included a number of first-hand accounts of the use of a dozen or so
psychedelics with enough detail for the reader to get a good feeling for the
set and setting. Woven in with this was the story of how the authors, Ann
and Alexander (Sasha) Shulgin (under the pseudonymous cloak of Alice
and Shura) met, loved, and eventually married. Like all good love stories,
a triangular suspense resolves when a German woman, with whom Shura
was initially involved, eventually fades into the permanent past tense. In
Book II of PIHKAL, the structures and syntheses of nearly 200 chemical
compounds were given in "recipe" form followed in most cases by brief
evaluations of their psychic or physical effects by self-experimentation on
the part of various anonymous volunteers. The compounds of PIHKAL's
Book II are all in the class known as phenethylamines, which explains the
acronym in the title: Phenethylamines I Have Known And Loved.
The present book is based on the same format, but the compounds
of its Part II now fall into that other structural class to which almost all the
remaining psychedelics belong, the tryptamines. TIHKAL stands for
Tryptamines I Have Known And Loved. In TIHKAL, the human love triangle
is no longer present, and so the first half of the book consists rather of a rich
smorgasbord of narratives and essays concerning the origins and uses of
psychedelics. Here the natural products chemist can enjoy a delightful
ramble through the astonishing diversity of creatures — from fungi and
frogs to flowers — which contain DMT or its chemical near cousins. The
psychopharmacologist can wonder if a clue to the auditory hallucinations
so typical of schizophrenia can be found in the unique activity of DIPT. A
Jungian psychotherapist will be challenged by Alice's stories of the gentle
synergism exercised by a skilled therapist and the entactogen MDMA. And,
sadly, the civil libertarian may find reason to fear that something much like

Our objective in presenting this volume was to produce an encyclopedia about a particular
group of psychedelic drugs, the phenethylamines. Phenethylamines are drugs with
profound mind-altering properties that we find both fascinating and rewarding. It is our
conviction that the varied potency, duration, and character of these many compounds
should be explored further, so that they may be rated as to efficacy and safety, and
become practical tools in the work of medical professionals and lay therapists, as well
as in the hands of spiritual guides and seekers. Their potential is great, but with great
benefit, there also may be great risks. In order to lay a solid foundation based on known
biochemistry, pharmacology, and toxicology, we have explored the available literature on
chemical, medical, and legal aspects of the known psychedelics, and attempted to present
that information so that future practitioners have a concise source of information to guide
further work.
